I set my camera on full auto so there's no need to jack with the settings then I wrap it in bubble wrap and cut out a hole for the lens.
I use a 64 gig card too so I don't have to change it.
I find with the extra bulk that I no longer need a tripod cause I can hold the camera super still!

On this note, it's probably also a grand idea to have a waterproof, removable bin or something for draining boots in your car.

I've made the mistake of leaving wet, gross boots in the trunk of my car and it seeped in, so now when I turn on the heater, my car smells like a giant fart.

Sound advice.

Take out an insurance policy on your camera through your renter/homeowner insurance. All the bags/covers/waterproof whatever won't help you if you tip a tripod or have it stolen out of your car. Its super cheap to do this my camera gear costs me less than $8 extra per month and I have no deductible on about $10K worth of gear.

Then just get a good lowepro/kata/pelican/molle/whatever DSLR bag that fits your gear and run with it. Most dedicated camera bags have some sort of tripod attachment built into them too.
